I haven’t watched anything to do with Destiel or beyond the second season and I can point at that. Right there. Because the joke doesn’t land without that awkward beat of Dean being insecure on the subject. Literally nothing happens there! It’s a nothing exchange! The scene doesn’t work unless Dean is queer in some way. I don’t mean gay, I don’t mean anything specific, but Dean has to be queer in order for that joke to land. If he’s a straight, cisgender guy with nothing else going on then why does he look awkward in that moment?
